The Study for Standardization of Transportation Container and Methods of the Human Tissues

Abstract
Purpose: This study was designed to set out a standardization of transportation container and methods which feasible for each tissue, processing and storage characteristics in order to distribute human tissue products more safely and sterilely.

Materials and Methods: Human tissue products were classified in terms of storage methods as freeze-dried storage tissue, refrigerated storage tissue, deep frozen storage tissue and ultra-low frozen storage tissue and the required coolants are as none, wet ice, dry ice and liquid nitrogen, respectively. The transportation containers such as cardboard container, styrofoam container, cryo shipper were being used. The experiments are reclassified according to seasons(summer, fall, winter), transportation distance and altitude. The safety test for the human tissues was performed by H-E stain, before and after each transportation for observing any changes of material. The temperature was measured with data logger (-200¢ªC~+800¢ªC).

Results: Assigned 12 and 6 hours staying time to Busan and Jeju city from Seoul on top of round trip time, the total is 0.96 day and 2.67 days, respectively. One way trip time from Branchburg (New Jersey, USA), Miami (Florida), Denver (Colorado) to Seoul was 2.45 days, 2.45 days, 2.63 days, respectively. A freeze-dried storage tissue was transported at room temperature (range: -11¢ªC~28¢ªC) in cardboard container. The result of a refrigerated storage tissue transportation is maintained refrigerate temperature(range: -1.4¢ªC~1.3¢ªC) for 2.5days (summer), 3.33 days (fall), and 3.70 days (winter) with 10.8 kg of wet ice in each styrofoam container. The result of a deep frozen storage tissue transportation is maintained frozen temperature (range: -72¢ªC~-77.6¢ªC) for 3.87 days, 5.9 days and 7.54 days in summer, fall and winter, respectively with 29.4 kg of dry ice in medium styrofoam container. The result of ultra-low frozen storage tissue transportation is maintained ultra low frozen temperature (range: -172¢ªC~-177¢ªC) for 3.3 days with 6 L(2.59 kg) liquid nitrogen in the cryo shipper.

Conclusion: A cardboard container was proper to maintained storage temperature during freeze-dried storage tissue transportation, a refrigerate storage tissue (raw material, rib cartilage, skin) transported in the medium styrofoam container with wet ice is proper to maintained refrigerate temperature range(10¢ªC or below). A deep frozen storage tissue in the medium styrofoam container with 29.4 kg of dry ice temperature was below -72¢ªC in any season for at least 3.87 days thus it is proper for transportation method. A ultra-low frozen storage tissue with 6 L of liquid nitrogen in the cryo shipper maintained temperature -172¢ªC for 3.3 days so it is proper as well. The gross histological result for each human tissue was normal by H-E stain.
